Automated MS/MS Optimization with Opt-MRM

What is Opt-MRM?
Opt-MRM was developed in collaboration with leading metabolism scientists at Amgen for high-throughput quantitation in N-in-one pharmacokinetics studies. Opt-MRM is a productivity tool that enables a user with a ThermoFinnigan Xcalibur-based TSQ to automatically determine optimal conditions for multiple reaction monitoring (MRM) experiments. You can perform multi-compound optimization with Opt-MRM and automatically determine the ideal MS and MS/MS conditions for each compound. Opt-MRM is ideal for quantitative studies requiring ultimate sensitivity, and/or for applications where multiple compounds in a mixture must be optimized (e.g., cassette dosing studies). Since Opt-MRM completely automates the process, you can determine the best conditions much more rapidly than if you were to perform the same steps manually.

Opt-MRM Features:
- Easy to setup and use - works through the familiar TSQ-GUIDE interface
- Includes tools for importing, building, and saving m/z optimization lists
- Optimizes MS and MS/MS conditions for up to 19 precursor ions
- Optimizes source voltages (tube lens and capillary) for each precursor ion in normal MS mode
- Capability to optimize positive and negative ions from the same m/z list
- In MS/MS mode, determines the "N" (up to 8) best fragment ions for each precursor ion (up to 19) in the user m/z list
- Plots collision energy breakdown curves for the "N" best fragment ions for each precursor ion
- Optimal MS/MS conditions (product ions and collision energies) automatically determined from the breakdown curves
- Saves optimal conditions for direct import into the TSQ experiment method editor
- For reference, automatically produces a log file and acquires product ion spectra using optimal conditions
- User selectable minimum product ion m/z, minimum neutral loss, "N" largest ions, intensity thresholds, and collision energy ranges for optimization
- Includes a full set of user documentation
Applications:
- Rapid, efficient method development for quantitative studies
- Optimization of MS/MS conditions for N-in-one pharmacokinetics studies
- Study effects of collision energy on compound fragmentation
- Use to determine energy-resolved collision conditions to improve selectivity in quantitative assays
System Requirements:
- ThermoFinnigan TSQ7000 or TSQ running on Xcalibur PC data system (except TSQ-Quantum)
- API-1 or API-2 ion source
- infusion pump
- Note: Opt-MRM is not currently compatible with older UNIX-based TSQ's.
