Witold,

The pressurized sample reservoir is a standard 100 mL pyrex media bottle, so you can have a sample loading bottle, a bottle of wash solvent, a bottle of eluent, and a bottle of "drano". You simply load your sample by placing your eppendorf tube in the bottom allowing pressure to load the sample onto the trap. After loading, unscrew the top and attach your wash reservoir and flush several uL's using the pressure. Now after you are done washing, you pull the silica above the liquid level and use the pressure to force all of the liquid from the trap. Your sample is now adsorbed to the trap on a nearly dry bed. You can then attach your eluent reservoir and elute your protein. The first uL that comes out contains your protein. You can add matrix to the eluent and elute directly onto a MALDI target. For this, I make up a satd. solution of matrix (sinapinic acid for proteins or ACHCA for peptides) in 70/30 ACN/H2O/0.1% TFA, centrifuge and draw off the top. Then dilute the satd. solution either 1:1 or 1:2 in 70/30 ACN/H2O/0.1% TFA. This can then be used as the eluent to elute samples directly onto the MALDI target.
