Novatia's Automated Biomolecule LC/MS System
A schematic of our automated LC/MS system for intact biomolecule analysis is shown below. The system is used routinely for the analysis of both oligonucleotides and proteins. Recently, we began offering this system as a fully-integrated solution for high-throughput analysis of oligonucleotides (see more info about the Oligo HTCS).

We have written custom control code for the LEAP/CTC-PAL autosampler that allows for rapid on-line pre-concentration and desalting of samples at the injector valve. We use the Michrom BioResources Paradigm MS4 as the HPLC system. The trapping column is a protein or peptide micro trap, also available from Michrom BioResources.
Sample Handling:
The autosampler first picks up a small volume of wash solvent followed by the sample volume followed by another volume of wash solvent. The PAL injector is rotated to LOAD so that sample is injected onto the trap. Biomolecules are retained on the trap, while salts, buffers, and other unretained solutes are diverted to waste. An additional volume of wash solvent is injected to ensure thorough desalting. For rapid analysis, the contents retained on the trap are eluted directly into the MS ion source with a strong mobile phase (e.g., 70% organic). For more detailed analysis an appropriate column is inserted after the valve and a gradient separation is performed. For protein analysis, the autosampler is programmed to inject a special cleaning solvent ("protein drano"), which eliminates sample carryover between injections.

Data Processing:
Once data acquisition is complete, data are automatically processed using our ProMass ESI deconvolution and web-based reporting software.
